Born to Indian parents in Uganda, Mamdani immigrated to the United States when he was a small child. His father, Mahmood Mamdani, was born in Uganda and is a professor at Columbia University. His mother, Mira Nair, is an award-winning filmmaker, as per a report.

Zohran Mamdani has been an outspoken opponent of the Israeli occupation government's policies toward the Palestinian people for a long time. He proposed a bill in 2023 to end the tax-exempt status of charities in New York that have connections to Israeli settlements that are in violation of international human rights law.

Mamdani has repeatedly distinguished between anti-Zionism and antisemitism, saying that antisemitism has no place in New York City.

However, Mamdani's support for Palestine and opposition to Israel have drawn criticism. Jewish Voice for Peace Action, a pro-Palestinian Jewish organization, has endorsed him. For his position, he was called antisemitic by leaders of groups like the Far Rockaway Jewish Alliance, the Metropolitan Council on Jewish Poverty, and the American Jewish Committee.
